Experimenting with #ThT stain on #spider webs to see if it successfully bonds to the beta chain #fibrin / #amyloid like components. I've applied the stain and naturally under 460nm webs fluoresce the expected green. I'm waiting for the sample to completely dry before differentiating with alcohol and then using a water based mount to finally disassociate the unbounded stain from the webs and other artefacts. All being so, we might be left with ThT stained #spidroins #science #biology #microscopy

Did a new Alzheimer's drug keep this patient's brain healthier for longer?

New drugs Leqembi and Kisunla may have a modest benefit, don't cure #Alzheimer's, have a host of side effects (including brain bleeds), are troublesome to use (require IV infusion or injection) and multiple brain scans and cost 10's of thousands to administer.

"I think it helped," says her husband, Ken Bell. "But I'm not sure."

That sort of uncertainty is common when it comes to #Leqembi and #Kisunla

Both drugs clear the brain of a substance called beta-#amyloid, which forms the sticky plaques associated with #Alzheimers.

Will this be the death knell for Alzheimer’s monoclonal antibody treatments 🤔

Preprint on #Alzheimers drug deaths ignites dispute among authors

“Infighting among a group of prominent Alzheimer’s disease researchers has led to the withdrawal of a preprint they co-authored, which suggested a new Alzheimer’s drug markedly increases the risk of death. One scientist involved in the work charges the senior author failed to seek the go-ahead from his co-authors before posting an edited version of the article to a preprint server.”

Secondary nucleation is increasing recognized as a dominant process in protein aggregation, and in particular also for pathogenic (disease causing) proteins. Still, many questions remain about how this process works, and what implications it has for pathogenic mechanisms. Here we show the unique power of HS-AFM to actually visualize this process directly. Check it out!

@CStamp @DemocracyMattersALot This will be important for many people already dealing with memory issues or with family members showing possible signs of #dementia, however it is still limited by lack of any real treatment options. The closing paragraph says, “For people without memory loss who are worried about genetic or familial #Alzheimers risk, blood tests are not yet recommended. #Amyloid buildup can begin decades before symptoms appear, and no preventive treatments are currently available apart from general guidelines about healthy eating and living.”

A central role for amyloid fibrin microclots in long COVID/PASC: origins and therapeutic implications ('22)

portlandpress.com/biochemj/art

> Although the symptoms of Long COVID are multifarious, we here argue that the ability of these fibrin amyloid microclots (fibrinaloids) to block up capillaries, and thus to limit the passage of red blood cells and hence O2 exchange, can actually underpin the majority of these symptoms. Consistent with this, in a preliminary report, it has been shown that suitable and closely monitored ‘triple’ anticoagulant therapy that leads to the removal of the microclots also removes the other symptoms.

Do we have #Alzheimer’s disease all wrong?

“As Science noted in its story on the retracted paper, scientists are still debating whether the #amyloid theory is viable. The skeptics cite the fraudulent research and lack of a genuine breakthrough; supporters can point to this new class of drugs including #donanemab that have led to some improvement in some patients.”

A long-awaited AlzForum's take on the news that ApoE4 homozygotism means in fact an AD pathology (though not necessarily dementia before death), alzforum.org/news/research-new

The comments-section gives a glimpse of some of the trenchlines regarding the treatment of dementia, but I guess the prevention-camp has only just been strirred up from their Sleeping Beauty's slumber.

I, as an example, still have only a 1 % measured risk of acquiring AD, according to some widely used screens, and no "preventable risk factors" except maybe age. Too bad, my genetic inheritance seems to have been enough to skew the Amyloid-Beta balance early on. Luckily, I was recruited to the aducanumab-study for four years (a precursor of lecanemab with more severe side-effects), and probably got some more years to be able to think things like these.
