For our lab's weekly journal club covering classic papers in immunology, we discussed Zheng and Flavell's discovery of GATA3 as a critical transcription factor for Th2 cell cytokine expression.

Before microarray or RNA-seq, there was cDNA subtraction by representational difference analysis.

2. There is a very honest description of scientists' motivations. We are all "epistemically sullied", motivated by both curiosity AND desire for recognition.

It is critical to acknowledge this as we seek to reenvisage publishing. Denying the fact (how it drives pursuit of JIF, CNS, h index, competition, etc.) won't help us. The key is to understand this and then think how the system can be improved so distortions (Goodhart's Law, etc.) are minimized.

1. There's a great explanation of the oft-cited Why Most Published Research Findings are False paper by John Ioannidis that explains why assumptions made about how scientists general hypotheses mean this is not correct.

Others have pointed this out but Carl and Jevin give a very clear explanation of why, which is important given how frequently and uncritically this work is cited in our sphere 2/3

Calling Bullshit by @ct_bergstrom is a fascinating read that brilliantly summarizes issues around data interpretation, various logical fallacies, phony graphs, and misleading statistics.

That alone makes it essential reading but two other things stood out for me that may interest sci comm/publishing types. 1/3

90% of cancers reactivate telomerase. In our new preprint, we reveal that tumors that fail to activate telomerase trigger a stronger immune response, leading to melanoma regression. Our work also sheds light on the association between telomerase gene mutations and the immune system and provide a mechanism to explore the success of immunotherapies.
